JUSTIFICATION: Despite clear indications of links between enteric microbiota and multiple human diseases, there is still no clear mechanism established, at least not to the level for which targeting gut microbiota for disease treatment can be reasonably successful, partly due to a lack of effective research tools. While many techniques exist for measuring effects of microflora on a host, and multiple animal models exist, the intense methodologies for maintaining gnotobiotic and knock-out models limits progress in the field. Investigations into the effects of regulation of host function by early microflora colonization has typically utilized gnotobiotic or disease models in mice. An inherent handicap of these models is a lack of natural phenotype and complications related to unintended physiologic changes. Probiotic studies in mice with pseudo-controlled enteric microflora have established some important relationships between the microbiome, inflammation, skeletal development, and obesity, to name a few. While these models provide critical information about interactions and symbiosis of the enteric microbiome and vertebrate animals, maintenance of controlled microbial environments, especially from birth, can be cumbersome, and expensive. Avian species provide a distinct opportunity, through prenatal in ovo inoculation of bacteria in order to evaluate early colonization, possibly in combination with enteric inflammation models, to combine both microbiome and immune function studies of these interactions and influences on multiple aspects of health and development. Direct inoculation of the amniotic fluid near hatch, which is swallowed by the embryo immediately prior to hatch, is easily accomplished with chicken embryos by in ovo injection. Recently, completed studies have shown that in ovo injection of chicken eggs at 18d of incubation can markedly influence recoverable bacteria from the GIT 48h after hatch (manuscript in preparation). These experiments did not include any attempts to decrease inoculation with indigenous microflora during the early postnatal period. We believe that modification and extension of this existing protocol can greatly enhance the ability to effectively study host-microbiome interactions and provide for a robust model towards understanding the role of specific pioneer colonizers of the intestinal tract of neonates. This research will contribute to scientific knowledge or public welfare.

Goals / Objectives
Bacterial communities living and colonizing in the gastrointestinal tract of animals outnumber total somatic cells of metazoans by an estimated 10-fold [1]. Today, the microbiome is recognized as the 'forgotten organ,' operating like an organ within the host and orchestrating numerous physiological and biological functions that have a profound impact on the balance between health and disease [2]. Essential colonization of these bacterial populations starts at birth/hatch, and is followed by progressive assembly of a complex and dynamic microbial society [3]. Under commercial conditions, millions of chickens and turkeys hatch in a hostile environment, and are exposed for several hours to heat stress and potential pathogenic bacteria in the hatcheries. Increased stress along with the potential abundance of pathogens in the hatching cabinet leads to ideal conditions for pathogen colonization. It is generally accepted that the natural route of transmission of zoonotic pathogens such as Salmonella, is fecal-oral [4]. However, published studies from our laboratory have also suggested that airborne transmission of Salmonella in poultry is possible [5]. This mode of infection could explain some clinical expression of relatively low-dose infectivity under field conditions in relation to the high oral challenge dose that is typically required for infection through the oral route in laboratory studies. This also supports previous studies demonstrating fan driven spread of Salmonella within the hatching cabinet and hatchery incubators [6]. Modern poultry production has separated the newly hatched chick from the hen and the complex microbiota that is maternally transmitted. Chicks are now born into the sanitized environment of the hatching cabinet where the first gut colonizers are often pathogens such as E. coli or Salmonella. Before hatching, the environment within the egg exists in a sterile state. In ovo application of probiotics may allow colonization of beneficial bacteria before the chick is ever exposed to pathogens within the hatching environment. In this research, multiple lactic acid bacteria (LAB) isolates will be screened for their impact on hatchability and their ability to colonize the gut after in ovo application. Hence, the objectives for this proposal are:To evaluate the effect of the in ovo administration of FloraMax®-B11 on MD vaccine herpesvirus of turkeys (HVT) protective efficacy, hatchability, microbiota composition, morphometric analysis, and SE infection in chickens. These results will provide preliminary data that can be used to leverage funding for investigation into the effect of GIT microbiome manipulation on immune function profiles. Overall, this model will provide a means by which to study effects of microflora on multiple health parameters, as well as population manipulation strategies towards favorable enteric microbe conditions that promote a symbiotic relationship.Incubation is a crucial step in a chicken's life cycle, and the environment in which an embryo develops can have lasting effects on the bird's well-being throughout its life. It has been documented that temperature, humidity, sound, and light can all impact development of an embryo. Stress hormones have been associated to increase intestinal permeability. Therefore, a second objective for this grant is to evaluate the effect that heat stress or cold stress has on inflammation and intestinal permeability and susceptibility to yolk sac infection and bacteremia. In these trials, hatchability, microbiota composition, and morphometric analysis will be also evaluated.3. To evaluate the administration of probiotic bacteria sprayed in hatching cabinets as an alternative to formaldehyde to sanitize and provide early colonization with beneficial bacteria to the hatching chickens. For these experiments, incidence of Salmonella, microbial composition in the gastrointestinal tract, metagenomics analysis, gut permeability, and serum biomarkers (citrulline, MUC2, IFN gamma, superoxide dismutase and total IgA) will be compared between chickens hatch in an environment where formaldehyde has been used versus spraying of probiotic bacteria.
Project Methods
PROCEDURES: Our central hypothesis that prenatal enteric inoculation of selected bacterial isolates, via amnionic injection prehatch, will manipulate pioneer colonization of the GIT and altered host GIT/immune responses will be tested by addressing the following:We will evaluate the effect of the in ovo administration of FloraMax®-B11 on MD vaccine herpesvirus of turkeys (HVT) protective efficacy, hatchability, microbiota composition, morphometric analysis, and SE infection in chickens. Potential interference with MD vaccine will be done incollaboration with Dr. Hans Chen at ADOL and will include effect on PFU and resistance to challenge in vaccinated birds. Microbiota composition will be performed by high throughput sequencing and description of relative abundance through the genus level in all cases. Morphometric analysis will be performed as previously described from our laboratory (Menconi et al., 2015).Incubation is a crucial step in a chicken's life cycle, and the environment in which an embryo develops can have lasting effects on the bird's well-being throughout its life. For these experiments, incidence of Salmonella, microbial composition in the gastrointestinal tract, metagenomics analysis, gut permeability, and serum biomarkers (citrulline, MUC2, IFN gamma, superoxide dismutase and total IgA will be compared between chickens hatch in an environment where formaldehyde has been used versus spraying of probiotic bacteria.While we have obtained significant differences in FITC-d intestinal permeability, we believe we can optimize the ELISA method to obtain more robust windows between control or treated chickens. Hence, one of our priorities will be working in the optimization technique. Ongoing studies are evaluating dosage, timing and the optimal MW of the dextran-FITC conjugate.The mucosal surfaces (gastro intestinal, respiratory, urogenital, ocular and inner ear) represent a very large exposure area to exogenous agents including microorganisms. As such, these anatomical sites are the main areas of interaction of the body with the external environment and thus with potential pathogens. The mucosal associated lymphoid tissue (MALT), differs fundamentally from Systemic immune responses in that: the major isotype in mucosal secretions is secretory, IgA and most of the antibody-producing cells and effector T cells occur in the MALT. Interestingly, the gut associated lymphoid tissue (GALT), as part of the MALT, is the largest immunological organ in the body, comprising 70-80 % of all immunoglobulin producing cells. GALT produces more secretory IgA (50-100 mg/kg body weight/day) than the total production of IgG in the body (30 mg/kg body weight/day)[21]. Hence, during this coming year, we will focus on an ELISA method for quantification total and specific IgA from different chicken MALT tissues.

Target Audience:Our target audience continutes to be the Poultry Industry and its needs. As regulations increase and antibiotics become less avaliable, we are working to find affordable and effacious products that meet the needs of poultry producers throughout the world. We have worked with numerous poultry integrators and major players in the poultry allied industry to try and meet these goals. Changes/Problems: Nothing Reported What opportunities for training and professional development has the project provided?This project assisted in the graduation of 1 PhD student during the last year, and has provided opportunities for one postdoctoral associate and one scientist from soft funding sources. Moreover, several undergraduate students have been exposed to our area of agricultural research, some may continue their studies following graduation. How have the results been disseminated to communities of interest?Journal article, presentations with abstracts and invited presentations internationally are listed. Numerous face to face meetings with industry leaders. What do you plan to do during the next reporting period to accomplish the goals?We will continue to work toward sustainable non-antibiotic solutions to ongoing and emerging poultry health issues.

How have the results been disseminated to communities of interest?Refereed publications, regional and national meetings, and individual meetings with numerous members of the U.S. poultry industry. What do you plan to do during the next reporting period to accomplish the goals?Progress is coming very quickly on this project. We anticipate several invention disclosures and approximately 12 refereed publications during the next year.

Impacts
What was accomplished under these goals? Several projects involving pre-natal or neonatal exposure of chicks to beneficial microflora were published, and intellectual property protection is being sought (Patents Pending). Two of these projects are under commercial development by multinational companies for use in broiler hatcheries, and turkey hatchery application is being investigated. Further progress of recombinant vaccine development was also reported in the citations. Major impacts include development of models for induction and measurement of leaky gut (including bacterial translocation) were completed. These models are currently being used for efforts to ameloriate the impact of enteric stressors on improving gut barrier function in broilers and turkeys.
