Goals / Objectives
Develop genome editing systems for maize based expression of CRISPR-Cas from RNA virus vectors.
Project Methods
We will develop maize infecting rhabdovirus-based virus vectors to deliver Cas nucleases and guide RNAs to maize using the virus¿ planthopper and leafhopper vectors. We will develop infectious rhabdovirus clones and will use these to stably express green fluorescent protein in maize. Subsequently, we will develop protocols for insect transmission of modified viruses expressing Cas9 with guide RNAs and other phenotype altering proteins to maize. We will assess the capability of the system in large-scale greenhouse experiments that will also evaluate the potential for viruses and expressed genes to move into non-target populations.
