Goals / Objectives
Confirm identification of USDA accessions of species in the genus Medicago section that includes secondary and tertiary wild relatives of alfalfa using DNA barcoding and ploidy level. Obtain relevant information about genetic diversity among accessions of crop wild relatives of alfalfa. Confirm identification of selected USDA accessions of species most closely related to M. truncatula and other species in a revised subsection Pachyspireae using DNA barcode sequence data. Provide a rough estimate of the proportion of accessions that might be misidentified within groups sampled. GRIN-Global: Add information for sampled accessions of Medicago. Contribute to student education and training with the goal of increasing understanding of crop wild relatives, molecular biology techniques and importance of study of crop wild relatives in agriculture.
Project Methods
Project attempts to delineate and potentially identify taxonomically mislabeled accessions within the temperate forage legume germplasm accessions focusing on species within the Medicago genus. This will be accomplished by screening a representative sample of accessions for their ploidy by using flow cytometry and by examining nuclear and organellar DNA sequences (¿DNA barcoding¿). Ploidy and sequence information will be compared to known true-to-type taxonomic samples and sequence information in NCBI¿s GenBank. Phylogenetic analysis will use standard software tools for sequence comparisons. Information generated will be analyzed and information on any potential mislabeling will be sent to the ARS Principal Investigator for correction. Accession-associated data generated will also be incorporated into the USDA-ARS National Plant Germplasm System¿s GRIN-Global database, into NCBI¿s GenBank databses as well as assembled into a peer-reviewed publication.
